Leave it on all the time. There are usually 10 settings; pick one (maybe start at 5) and see how long the tray takes to fill up. Then adjust the setting as necessary so that the humidity level in the basement is comfortable. When you know roughly how long it takes to fill the tray, you'll know your schedule for emptying it. It will shut off automatically when it's full.

Sears has humidifiers on sale now. I just purchased a 65-pint/day humidifier for $179.99. It has a humidistat so you can regulate the humidity much easier. Completely automatic. Just set it to 45 or 50% and walk away. It took about 48 hours to drop the humidity from 73% to 43% in my 1000 sq. ft. basement.

Many dehumidifiers do not have a guage. I bought an inexpensive temperature/humidity gauge to hang on the wall opposite the dehumdifier. I set mine to run until the guage reaches 70%. That level seems to work in my scenerio.
I also drilled a hole in the foundation floor to run a hose from the dehumidifier. It's a lot easier than emptying the container periodically. In my case, drainage under the floor was good, and I have not had a problem with water seeping back in.
