You have two different issues.
Apple iOS devices (iPhone/iPad) will only print to AirPrint enabled devices, which many printers aren't. Apple provides a list, but yours isn't listed.
There are some applications you can run on your Windows computer that function as an AirPrint 'controller'. Your iPhone will print to this controller, then to the printer. The downside is that your Windows computer needs to be on and connected for it to work. I've seen some of these software packages out there, but I haven't used them, so can't recommend any.
As for your XP machine, I think you need to install the Brother drivers to enable the wireless functionality.

I've had good success with Brother printers using the full driver install package (the first on the list). You may be able to pick and choose, but I usually don't bother. I've found that Brother doesn't add a lot of crap like other printer drivers do.

Downloaded Brother™ iPrint&Scan Apple® app said printing, nothing printed. How should everything be setup? Printer hard wired to router, WiFi, or to desktop that's not WiFi?

You'll need to connect the printer to your router either using the ethernet cable or Wi-Fi.
Here's the quick start guide to setting up your printer: Brother: HL-2270DW: Downloads: United States
And the link to download the drivers for your Operating System: Brother: HL-2270DW: Downloads: United States

Start with the Windows full driver package and get your XP desktop working first. You'll need the Brother Windows XP drivers to get it working.
After you get the XP machine working, then you can focus on the Apple drivers.

I had a Brother MFC-495CW on Win 7 Pro that would not print wireless, no problem over the wire, and it was kickin my a55. Nothing worked, not even HP's forum. Then a one line post buried in a printer forum some where mentioned a tiny file named ntprint.inf.
