Can I bypass BR air return and install a vent?

I want to put a 1st floor doorway where a 2ND floor BR air return wall channel to the basement is. Can I place a vent above the new 1st floor doorway to vent the 2nd BR? Will this still affect pressure in 2ND floor BR or hurt resale? Thanks!

I'll also mention that I'm planning to add a vent to the same return duct in the basement to keep the air flow to the AH.

As long as you have a way for the air to get from 2nd story to AHU you will be fine

All my BRs on the 2nd floor have returns to the AHU. What I want to do is by pass the return for one of the BR and put in a vent instead.
