Do you add the boots before or after drywall?
i imagine it's difficult to seal the vent around the boot with tape if the drywall is there and you just have a rectangular cut out to work through?
But the boots don't mount easily on studs...

The boot has to be installed before sheetrock. The duct boot can only be installed while the wall cavity is open and you have access to it. Once the sheetrock is up you don't have access inside the wall to do much of anything.

For small sheetrock jobs I just go at it. For bigger jobs however I spend a little time before hanging any sheetrock to mark the location of every outlet/switch box and duct. I mark on the floor with an elevation to the center of the box. Then if you missed a cutout you can look at the floor and see where to find the hidden switch box or HVAC register. And after hanging the rock I look around the room to make sure I have a cutout for each of my marks.
