20A-125/250V wiring help

I have a generator that has a 250 volt connection. I have been using a one of the 120v connections to run power through my box trailer for lights and power. How do I go about wiring the 250 up to supply the trailer. I have run regular house type wiring through the trailer within plastic conduit that's three wire. This plug is four, what do I do to convert?

I'm not really sure what you are asking.
125/250v is basically two 120v circuits sharing a neutral, and also providing 240v.

What is the lead in connection to the trailer like? Is it also 4-wire?

No - it's regular household 3 wire

You need to wire the circuits from the trailer to a sub panel then feed the sub panel from the generator.

This is a bit much for a DIY project, but if you are handy, it is not impossible.
