masonite siding

I have masonite siding that is needing to be repainted. Can this type of siding be sanded to smooth out rough spots prior to painting?

The color on Masonite siding was put on there at the factory and baked in the surface. You can surely repair it and repaint it, but not in pieces, since you will never get the finish that came on it originally. Long story short, you will have to paint the entire house, or at least a side to get a good appearance.

You can sand the masonite but go easy with it. Masonite can deteriate quickly when exposed to weather [if the factory or paint finish is missing] It is best to use an oil base primer after sanding, top coat with your choice of latex house paint.
