Squeeky floor - strange design

I have a 40-year old house with a second floor that squeeks. The floor joists are 2X12's - 2X4's were placed on top of the joists at a 90-degree angle (flat), but they are not continuous, they are spaced 1-foot on center. Normal 3/4-inch plywood was placed on top of the 2X4's. How do I remedy the squeek since I have two layers on top of the floor joists, not the normal single layer of plywood? Would screws do the job, maybe spaced every 6-inches?
