2 separate a/c systems

Im moving into a ranch home with a finished basement of about 2400 sq. feet (located in the downstate NY area) which has 2 separate central air conditioning systems (an extension was added with the second a/c but it is configured to cool other parts of the home) of about 2 tons each. Is this setup more efficient than having one 4 ton unit?

Yes and no

You need to know the efficiency of the units (SEER rating), how they were installed/maintained and so on.
IMHO, with two system you have more control (get some programmable thermostats) and will have a backup when/if one breaks.

Most larger houses now are going with 2 or more systems.

I'm having a mini-split ductless AC system put in next month for my upstairs
