Toilet tank will not refill after flushing

When I flush my toilet, it will not refill unless I loosen the screw on top of the tower inside the tank. Is there a bad valve or do I need to replace the innards?

Tower? are you talking about the fill valve?

I would replace the fill valve is this is what are are talking about.

Lost here. You say the tank doesn't fill?
Then you do need to replace the fill valve.

If the bowl doesn't fill, there should be a tube running from the fill valve to the overflow tube. This is where the water comes from to fill the bowl.

If that tube is leaking or missing, replace it. Better yet, replace the complete fill valve assembly and this should correct all the problems.
