tub faucet handles

I have a dual handle tub faucet with shower diverter in the
middle of the hot and cold handles. I want to replace all
3 handles. Do I have turn off the water supply to replace
the handles? If so, where do I find the water supply valve?

No, you do not have to cut-off the water just to replace the handles.
However, you do have to cut-off the water to remove faucet valve assemblies that the handles are attached to, depending upon what you meant by "handles".
You may have individual cut-offs for the hot and cold valves, which would have to be accessed from the wall behind the tub.
It would be simpler (rather than cutting into the wall if you don't have an access panel) to just cut-off the main water valve to your house. (Cut the power off to your water heater, too.)
