First thing to try is lift/push down and pull/push on door while trying to turn it. If the door is binding due to swelling, sagging or friction on the bolt, this will alleviate it.

If that is not it then there may be a mechanical failure, usually in the latch or tailpiece. Remove the inside thumbturn/plate. You can then remove the exterior cylinder assembly. If the tailpiece is still connected to the cylinder then it is probably a broken bolt. You should be able to insert a screwdriver into the bolt where the tailpiece was-a working bolt will turn easily with the screwdriver. Check for any wood chips or other debris that may have fallen in to the bolt works. If it still does not turn, use a vise grip or pliers on the screwdriver for extra turning force to retract. If still stuck, you may have to remove the bolt piece by piece.
