Deadbolt handle cannot be turned from inside
My door deadbolt handle cannot be turned from inside so the door cannot be opened. Please see attached photo.
How to fix this problem?

Since you have access, just remove the two screws. The inner and outer sections should come apart, then you can see if it's the lock mechanism or the strike.

Push the door in with your hand and turn the thumb turn CLOCKWISE. If that doesn't work, try to pull the door open a little or wiggle it in and out while you turn the thumb turn clockwise. The way the thumb turn doesn't sit level may be an indication that the latch may be toast.

You need to take the two black screws out to remove the inner plate and the outer cylinder. That should leave the latch in the door.

I should have said it APPEARS to be backwards. Usually the raised portion of that plate drops into the 2" hole behind, and helps keep everything aligned inside and out. If it's slipped, it could put a bind on the latch.

This problem only occurred yesterday suddenly.
The left screw was loose. I tightened it but this hard-to-turn problem occurred so I posted this question.

So do you have the door open or not? If it is just difficult to turn, disassemble it completely, take the latch out and lubricate it with some WD-40 or similar. See if that helps. See if the gold plate is turned the correct way, if it isn't, correct it as that will help with alignment. If none of the above helps, get a different latch. They are like $5.

Currently I discover the optimum tightness of the screws - not too loose nor too tight and push on the door from inside, then the door deadbolt handle can be turned.
I also put some Graphite Lubricant in the keyhole and the door key is easier to turn now.
What else can be done?

If the hole behind it is 2" then yes, its backward. Thought I described how it works in post #7.
If you have to push the door from the inside, you likely need to grind a little off the inside edge of the deadbolt strike plate... or maybe remove it, fill the holes, and move the strike plate 1/8" closer to the inside. If it turns fine when the door is open then its simply binding on the strike plate. That's what I was alluding to in post #3.
