hole measurement

I'm installing wide pine a a small bathroom in my kitchen area. One board will require cutouts for the sink and toilet. What would be the best technique to accurately measure the placement of all cuts. I was planning on cutting them with hole saws and don't want any mistakes as this stuff is not cheap.

Thanks

I usually avoid measurements in this situation. When my floor reaches that point, I mark the locations on the board, in the exact position it will occupy.
