necessary to apply plumbers putty under bathroom sink flange?
It seems unnecessary to apply putty between the basin and the drain flange. The reason is because just below that is the overflow chamber. It seems to me that the basin officially seals at the lower rubber seal/nut. thoughts?

I have sometimes had difficulty getting the rubber washer to seal completely around the threads. Probably OK to get a few drips from the rare overflow use but a problem for the daily use of the main drain so I use plumbers putty. You might get lucky and not need it but putty is so cheap and easy I just go ahead and use it.

Thanks Dane - just so I'm clear, you and I are talking about the sink drain flange on the top side of the basin, not the underside correct?

I was going to use a thin bead of silicone around the bath tub drain flange,
should putty be used in place of the silicone?
I will add the Teflon paste to the mix too.
